喜炎平注射液对腺病毒感染Hep-2细胞Bcl-2/Bax蛋白表达的影响

Effect of Xiyanping injection on expression of Bcl-2/Bax protein in adenovirus-infected Hep-2 cells

摘要:
目的:观察喜炎平注射液对腺病毒感染Hep-2细胞Bcl-2/Bax蛋白表达的影响,探讨喜炎平注射液体外抗病毒机制。
方法:用腺病毒感染Hep-2细胞,分别予喜炎平高、中、低剂量组及利巴韦林干预,采用免疫组织化学法检测各组Hep-2细胞的Bcl-2、Bax蛋白表达情况。结果:模型组Bax蛋白表达显著高于正常组,差异有统计学意义(P<0.05);喜炎平各剂量组及利巴韦林对照组Bax蛋白表达均低于模型组,其中喜炎平高剂量组、利巴韦林对照组Bax蛋白表达显著低于模型组,差异有统计学意义(P<0.05)。各组Bcl-2蛋白表达比较差异均无统计学意义(P>0.05)。
结论:喜炎平注射液通过抑制Bax蛋白表达的上调发挥体外抗腺病毒的作用,高剂量组优于中低剂量组。

Abstract:
Objective:To explore mechanism of antiviral mechanism of Xiyanping injection in vitroby invstigating influence of  Xiyanping injection on expression of Bcl-2/Bax protein in adenovirus-infected Hep-2 cells.
Methods:The Hep-2 cells were infected with adenovirus, which were interfered with high, medium and low dose of Xiyanping injection and ribavirin. Then expressions of Bcl-2/Bax protein were measured by immunohistochemistry.
Results:The expressions of Bax protein in model group were significantly higher than normal groups(P<0.05).The expression of Bax protein in all Xiyangping groups and ribavirin control group was lower than that of model group, and it was much lower in the high-dose Xiyanping group and ribavirin control group than in the model group(P<0.05). The expressions of Bal-2 protein in different groups had no significant difference(P>0.05).
ConclusionXiyanping injection can kill ADV by suppressing the expressions of Bax protein. The high-dose Xiyanping injection is better than others.
