清肺通络膏对呼吸道合胞病毒感染大鼠肺组织PI3K-AKT-NFκB信号通路的影响

Effect of Qingfei Tongluo ointment on the PI3K/Akt/NF-kappaB signaling in rats infected with RSV

摘要:
目的:观察清肺通络膏对呼吸道合胞病毒(RSV)感染大鼠肺组织PI3K、AKT1/2、NF-κB p65蛋白表达的影响,探讨清肺通络膏对PI3K/AKT/NF-κB信号通路的调控机制。
方法:将30只SD大鼠随机分为正常组、模型组、治疗组各10只,模型组、治疗组均给予RSV滴鼻接种3 d,治疗组于接毒第1天予清肺通络膏背部贴敷,连续7 d。于造模第7天后取大鼠肺组织,运用免疫组织化学染色法检测各组大鼠肺组织中PI3K、AKT1/2、NF-κB p65蛋白的表达。
结果:与正常组比较,模型组大鼠肺组织PI3K、AKT1/2、NF-κB p65蛋白表达增多,差异有统计学意义(P<0.05)。治疗组大鼠肺组织PI3K、AKT1/2、NF-κB p65蛋白表达均低于模型组,差异有统计学意义(P<0.05)。
结论:RSV感染大鼠时激活了PI3K/AKT/NF-κB信号通路,清肺通络膏能够抑制PI3K、AKT1/2、NF-κB p65蛋白的表达,可能是清肺通络膏干预RSV肺炎的机制之一。

Abstract:
Objective:To observe the effect and the mechanism of Qingfei Tongluo ointment on the expression of PI3K,AKT1/2 and NF-κB p65 in rats infected with RSV.
Methods:Totally 30 rats were randomized into 3 groups,namely,normal group,model group,treatment group,with 10 rats in each.The rats of the last two groups were made into RSV infection models.For treatment group Qingfei Tongluo ointment was given at the first day of the experiment,once daily for seven consecutive days.Examine the levels of PI3K,AKT1/2 and NF-κB p65 in lung tissue,using the method of immunohistochemistry.
Results:The result of PI3K,AKT1/2 and NF-κB p65 expression in lung tissue showed that:compared with the normal group,the model group had significant difference(P<005);compared with the model group,the treatment group had significant difference(P<005).
Conclusion:Qingfei Tongluo ointment is an effective drug for external use in RSV infection;its action mechanism may be through adjusting the PI3K/Akt/NF-κB signaling.
