川崎病患儿治疗前后外周血单核细胞P22phox表达的变化及其临床意义

摘要:
目的
探讨川崎病急性期患儿治疗前后外周血单核细胞中P22phox mRNA表达的变化及其临床意义。
方法
2015年10月至2016年6月川北医学院附属医院儿科收治住院的川崎病患儿40例,其中有冠脉损害者15例,无冠脉损害者25例。同期选择本院小儿外科择期手术患儿20例为正常组。采取荧光实时定量RT-PCR法分别测定40例川崎病患儿治疗前后和正常组儿童外周血单核细胞中P22phox mRNA表达的水平。
结果
川崎病组治疗前后P22phox mRNA的表达比较差异有统计学意义(P<0.05);川崎病组治疗前与正常组P22phox mRNA的表达比较差异有统计学意义(P<0.01);川崎病组治疗后与正常组P22phox mRNA的表达比较差异无统计学意义(P>0.05)。治疗前有无冠脉损害、治疗后有无冠脉损害、治疗前后有冠脉损害以及治疗前后无冠脉损害P22phox mRNA表达的比较差异均无统计学意义(P>0.05)。
结论
P22phox可能参与川崎病急性期的发病机制。

Abstract:
objective:
To investigate the changes and clinical significance of the expression of P22phox mRNA of peripheral blood mononuclear cells(PBMCs) in children with Kawasaki disease(KD) before and after treatment.
Methods:
Totally 40 Kawasakai disease patients admitted to Pediatric Department of Northern Sichuan Medical College Affiliated Hospital from October 2015 to June 2016(15 patients with coronary artery lesions,25 patients without coronary artery lesions) who received IVIG treatment were selected in the study, and 20 age-matched healthy children undergoing elective surgery in the Pediatric Surgery of this hospital were selected as the control group. The expression of P22phox mRNA of the peripheral blood mononuclear cells(PBMCs) were measured using real-time polymerase chain reaction(RT-PCT) in the two groups before and after treatment.
Results:
In the treatment group, there was statistical difference in P22phox mRNA expression before and after treatment(P<0.05); the expression of P22phox mRNA had statistical difference between treatment group before treatment and the control group(P<0.01);there was no statistical difference in P22phox mRNA expression between treatment group after treatment and the control group(P>0.05). The expression of P22phox mRNA was not statistically different in the following cases: with or without coronary lesion before treatment with or without coronary lesion after treatment, with coronary lesion before or after treatment, and without coronary lesion before or after treatment(P>0.05).
Conclusion:P22phox may be involved in the pathogenesis of Kawasaki disease at acute stage.
