甲型流感病毒FM1株诱导感染不同鼠龄小鼠肺炎模型比较

Comparison of pneumonia mouse models of different ages with infection induced by influenza A virus FM1 strains

摘要: 目的:评价昆明种小鼠哪一鼠龄段为甲型流感病毒FM1株最佳模型。方法:150只清洁级昆明种小鼠,雌雄各半,鼠龄均等,分为幼龄模型组(40只)、成龄模型组(40只)、老龄模型组(40只)、幼龄正常组(10只)、成龄正常组(10只)、老龄正常组(10只),观察小鼠状态,分别于感染3、5、7 d,检测其肺湿质量、肺组织肉眼病理评分、肺组织病理切片以及肺组织FQ-PCR病毒载量。结果:幼龄模型组14 d内死亡最多,整体状态最差,肺部病变最为严重,肺组织病毒载量呈递增趋势。成龄模型组14 d内死亡最少,整体状态最好,肺部病变少,肺组织病毒载量无复制的现象。老龄模型组整体状态在第3天出现肺组织病变,但之后有恢复的趋势,第5天后,病毒载量呈下降趋势。结论:幼龄昆明种小鼠为甲型流感病毒最佳理想模型。

Abstract: Objective:To evaluate which rat age segment of Kunming mice is the best model of influenza A virus FM1 strains. Methods:A total 150 Kunming mice of clean grade, half male and half female, with equal age, were randomly divided into young model group (40 rats), mature model group (40 rats), aged model group (40 rats), young normal group (10 rats), mature normal group (10 rats), and aged normal group (10 rats). The mice were observed, and respectively at the end of 3,5 and 7 days of infection were detected lung wet weight;lung tissue pathology score with naked eyes, lung histopathology and lung tissue FQ-PCR viral load testing were also performed.Results:In the young model group, the highest death occurred in 14 days, the overall state was the worst with most severe lung disease, and viral load in lung tissue showed an increasing trend. In mature model group, the death was the least in 14 days, the overall state was the best, basically there was no lung disease, and lung tissue viral load had no copying phenomenon. In aged rats, the overall state showed lung tissue lesions in the first three days, but there was a tendency to recover and after 5 days, viral load tended to decline.Conclusion:The young mice are the idealest model of influenza A virus.
