中药肾乐方对微小病变肾病大鼠血清、肾组织内皮素干预作用的研究#br#

摘要: 目的进一步探讨中药肾乐方的肾脏保护作用机制。
方法将48只SD大鼠适应性喂养1周后,随机分为6组:正常组、模型对照组、强的松组、中药低、中、高剂量组(中药肾乐方治疗)。采用表柔比星尾静脉注射法复制微小病变肾病(MCNS)大鼠模型,分别给予不同剂量肾乐方药液、强的松混悬液。测定大鼠24 h尿蛋白定量、血清白蛋白、总胆固醇、三酰甘油、尿素氮、肌酐、血清及肾组织内皮素(ET)的水平,并进行电镜下肾脏组织形态学观察。
结果与正常组比较,模型对照组血清、肾组织ET质量浓度均有不同程度增高(P<0.05~0.01)。肾乐方可减少MCNS大鼠尿蛋白的排泄量,改善血清生化指标,降低血清及肾组织ET质量浓度(P<0.05~0.01),修复肾小球上皮细胞足突的结构。
结论MCNS大鼠ET质量浓度升高,肾乐方对MCNS大鼠有明显疗效,并能抑制其增高的ET表达,从而减轻肾脏病理损害,缓解肾病。

Abstract: ObjectiveTo investigate the effects of Shenlefang in MCNS rats.MethodsA total of 48 MCNS model rats were induced by ADR venoclysis through caudal vein.The rats were randomly divided into six groups:the normal control group,the model control group,the positive control group,the lowdosage of Chinese herbs group,the middledosage of Chinese herbs group,the highdosage of Chinese herbs group.Different treatment were given to the six groups.We observed the amount of protein in urine,determined the levels of Alb,TC,TG,BUN,Scr and ET in serum and ET in kidney,and observed morphologic change in kidneys.ResultsCompared with the normal group,the levels of ET in serum and kidney tissue of rats in the model groups were increased(P<0.05~0.01).Shenlefang could decrease the amount of protein in urine and the TC,TG,BUN,Scr and ET in serum or kidney(P<0.05~0.01),and repair injured foot process of glomerule epithelial cell.ConclusionET may be concerned with the pathogenesis of MCNS.Shenlefang has obvious protective effect on MCNS rats,inhibit the ET expression in serum and kidney,and prevent the further development of MCNS,hence reduce the pathologic damage to kidney and alleviate kidney disease.
