Expression of ephrinB2 and EphB4 in a neonatal rat model of periventricular white matter damage resulting from hypoxia-ischemia

Abstract:
Objective: To evaluate microvessel density (MVD) and the expression of ephrinB2 and its receptor EphB4 in a hypoxia-ischemia induced PWMD rat model.
Methods: Postnatal day 3 Wister rats(n=60) were randomly divided into hypoxia ischemia group(group HI),sham operation group(control group),with 30 rats in each group.The HI group underwent permanent ligation of the right common carotid artery then they were exposed to hypoxia (94% N2+6% O2) for 4 hours.For the control group rats,the same surgery was performed except for ligation and exposure to hypoxia.MVD and levels of ephrinB2 and EphB4 were determined at postnatal day 7.
Results: Compared with control group,MVD,ephrinB2 and EphB4 levels were higher in the brains of hypoxic-ischemic rats.Similar percentages of vessels expressed ephrinB2 and EphB4 in control group,but expression of ephrinB2 was greater in brains injured by hypoxia-ischemia.
Conclusion: Following hypoxic-ischemic injury to the rat brain,there were angiogenesis and more arterioles than venules are acquired.
