清热解毒、化瘀通络方对系膜增生性肾炎大鼠肾组织肿瘤坏死因子α、内皮素1的影响

〗The effect of prescriptions of Qingre Jiedu and Huayu Tongluo on TNFα and ET1 in MsPGN model rats

摘要: 目的观察清热解毒、化瘀通络全方及其拆方组对系膜增生性肾炎大鼠肾组织肿瘤坏死因子α(TNFα)、内皮素1(ET1)的影响,以探讨清热解毒、化瘀通络全方及其拆方的作用环节。
方法本实验以雄性SD大鼠为研究对象,以隔日牛血清白蛋白灌胃并尾静脉注射葡萄球菌肠毒素B及皮下分点注射完全氟氏佐剂和不完全氟氏佐剂的方法复制系膜增生性肾炎模型。观察各组实验大鼠肾小球细胞外基质增生情况;免疫组织化学法检测肾组织中TNFα及ET1的表达并进行半定量分析。
结果复制系膜增生性肾炎动物模型,光镜下肾小球轻〖CD*2〗中度系膜增生。模型组和各观察组系膜增生性肾炎实验大鼠肾组织TNFα,ET1表达均有显著增高(P<0.01);在降低肾组织TNFα和ET1表达方面,全方组能显著降低两者的表达水平(P<0.01),两个拆方组各有侧重,清热解毒组偏重于降低肾组织TNFα表达,而化瘀通络组偏重于降低肾组织ET1表达。
结论清热解毒、化瘀通络多法合用在系膜增生性肾小球肾炎的治疗中较单一疗法更为有效,可从不同作用环节减轻肾脏病理损伤。

Abstract: Objective〖WT5"BZ〗Observe the effects on TNFα and ET1 expression in renal tissues of rats with mesangial proliferative glomerulonephritis(MsPGN) by Chinese herbs with functions of both the heatclearing and detoxification(HD,Qingre Jiedu),and the removing of blood stasis and channel obstruction(RBSCO,Huayu Tongluo),or each of them,in order to explore mechanism of the Chinese herbs.
〖WT5"HZ〗Methods〖WT5"BZ〗Male SD rats were included as the study object.The rat models of MsPGN were made by taking orally and timecontrolled vein injecting boving serum albumin(BSA),and vein injecting staphylococcus enterotoxin B(SEB),and spot of division of hypodermic injecting complete Freund's adjuvant and not complete Freund's adjuvant.The proliferation of glomerular mesangial cell(MC) and glomerular extracellular matrix(ECM) was studied by light microscope on rat models of MsPGN,who were treated by combined HD with RBSCO,and by HD,or by RBSCO,respectively.TNFα and ET1 expression in renal tissues was detected by immunohistochemical method,and quantitative analysis was made.
〖WT5"HZ〗Results〖WT5"BZ〗In the successful mesangial proliferative nephritis models,glomerular mesangial proliferation was mild to moderate,and TNFα and ET1 expression in renal tissues increased significantly(P<0.01).The treatment of combination of HD with RBSCO could significantly reduce TNFα and ET1 expression levels in the renal tissues.HD or RBSCO respectively might have different action mechanism,because HD group was emphased on reducing TNFα expression(P<0.01),while RBSCO Group was emphased on reducing ET1 expression(P<0.01).
〖WT5"HZ〗Conclusions〖WT5"BZ〗The treatment of combination of HD with RBSCO for MsPGN is more effective than that of single HD or RBSCO,and it can reduce the renal pathological damage through different treatment mechanisms.
