中药二步序贯治疗对哮喘小鼠肺组织GATA3表达及气道炎症的影响#br#

摘要: 目的探讨二步序贯治疗即中药人参五味子汤加小青龙汤对哮喘小鼠肺组织GATA3表达及气道炎症的影响。
方法30只SPF级雌性小鼠被随机分为正常对照组、哮喘模型组、哮喘模型胃饲中药组,每组10只。以卵清蛋白腹腔注射并雾化吸入制备小鼠哮喘模型,并先后予以人参五味子汤加小青龙汤对哮喘小鼠进行治疗。处死后ELISA检测各组小鼠血清中卵蛋白干粉剂(OVA)IgE水平;肺泡灌洗液(BALF)测定细胞总数及各类炎性细胞数目;HE染色观察肺组织病理变化,免疫组化测定肺组织中GATA3表达水平。
结果哮喘模型胃饲中药组血清OVAIgE水平、GATA3表达水平较哮喘模型组明显降低,较正常对照组升高。哮喘模型胃饲中药组BALF中白细胞总数、嗜酸性粒细胞比例、淋巴细胞比例较哮喘模型组降低,较正常对照组升高;而吞噬细胞比例较哮喘模型组升高,较正常对照组降低。
结论人参五味子汤加小青龙汤可能通过影响GATA3表达进而调节免疫系统,从而控制哮喘小鼠气道炎症,对其治疗起重要作用。

Abstract: ObjectiveTo study the effect of Xiaoqinglong decoction and GinsengSchisandrae decoction on the expression of GATA3 and airway inflammation in lungs of mice with asthma.MethodsThirty mice were randomly divided into three groups(n=10,in each group):normal control group,model group,Xiaoqinglong decoction and GinsengSchisandrae decoction group.The mouse group of asthma were made by the egg albumen sensitization method and treated early with Xiaoqinglong decoction and GinsengSchisandrae decoction.OVAIgE in the serum of mice were detected with enzymelinked immunosorbent assay (ELISA).The total number of cells in the BALF and the classification of cells were tested.The pathology of lung tissue was examined by HE.GATA3 in the lung tissue was examined with immunohistochemical method.ResultsThe OVAIgE and GATA3 were decreased in the Xiaoqinglong decoction and GinsengSchisandrae decoction group compared with the model group and was increased compared with the normal control group.The total cells and the percentage of eosinophils and lymphocytes in BALF in Xiaoqinglong decoction and GinsengSchisandrae decoction group were decreased compared with those in model group and increased compared with those in normal control group(all P<0.01).The percentage of phagocyte in BALF in Xiaoqinglong decoction and GinsengSchisandrae decoction group was increased compared with those in model group and decreased compared with those in normal control group(P<0.01).ConclusionXiaoqinglong decoction and GinsengSchisandrae decoction may regulate immune system by influencing the expression of GATA3,which plays an important role in treating asthmatic mice.
