Effect of Qingfei Tongluo ointment on the expression of ERK/NF-κB p65 protein in the lung tissues of rats with influenza virus pneumonia

Abstract:
Objective:To investigate the mechanism of Qingfei Tongluo ointment in regulating the EPK/NF-κB p65 signal pathway in the lung tissues of rats with influenza virus pneumonia.

Methods:Thirty Wister rats were randomly divided into 3 groups: normal group(A), model group(B) and Qingfei Tongluo group(C), 10 rats in each group. Influenza virus FM1 strain was inoculated through nose to induce pneumonia in young Wister rats, in group B and C. After infection, Qingfei Tongluo ointment was applied to the projected area of lungs on the back of rats for treatment. The general state of all the rats were observed, including the amount of food and water taken, hair,exercise, mental state and breathing frequency. The morphology of lung tissues was observed under light microscope. The expression of NF-κB p65 and pERK was detected by immunohistochemical assay.
Results:Compared with group A, the expression of p-ERK and NF-κB p65 protein in the lung tissues of rats in group B was significantly increased, and the difference was statistical(P<0.05);the expression of p-ERK and NF-κB p65 in group C was significantly reduced compared with group B, and there was statistical difference(P<0.05).
Conclusion:Qingfei Tongluo ointment can reduce the expression NF-κB p65 through inhibiting the phosphorylation of ERK, thus alleviating the pathological damage of lung tissues and achieving the treatment effect.
