Role of miRNAs in the model of asthma in mice

Abstract: Objective:To investigate the differential expression of miRNAs in asthma model of mice.
Methods:BALB/c mice were randomly divided into normal control group, asthma group(ovalbumin treatment group) and treatment group(dexamethasone treatment group), 8 mice in each group. Eosinophil(EOS), neutrophil(NEU), macrophage(MAC) and lymphocyte(LYM) in BALF of each group were detected by hematology analyzer, and the expression of IL-4,IL-6 and IL-13 in BALF of each group were detected by ELISA. miRNAs microarray was used to analyze the differential expression of miRNAs in different groups of lung tissues; the correlation between miR-455 and EOS,NEU,MAC,LYM,IL-4,IL-6 and IL-13 was statistically analyzed.
Results:The expression of EOS,NEU,MAC,LYM,IL-4,IL-6 and IL-13  in the asthma group was significantly higher than that in the control group, and the expression of various factors was downregulated in treatment group compared with asthma group. The expression of multiple miRNAs in asthma group changed,and miR-455 was up-regulated significantly in asthma group. miR-455 was positively correlated with the expression of EOS,NEU,MAC,LYM,IL-4,IL-6 and IL-13.
Conclusion:miR-455 is significantly up-regulated in ovalbumin-induced asthma model of mice.
