    Experimental research into the effect of Qingzao Jiufei decoction on rats infected with mycoplasma pneumoniae

    【Abstract】Objective:To observe the effect and the mechanism of Qingzao Jiufei decoction on pathological inflammation of lung tissue and bronchoalveolar lavage fluid (BALF) in rats infected with mycoplasma pneumoniae (MP).
    Methods:Totally 90 rats were randomized into 6 groups, namely, normal group (group A), model group (group B), Qingzao Jiufei highdose group (group C), Qingzao Jiufei mediumdose group (group D), Qingzao Jiufei lowdose group (group E) and roxithromycin group (group F) ,with 15 rats in each. The rats of the last five groups were made into MP infection models. The same volume of drugs or normal saline was given to each group by introgastric administration respectively. The pathological inflammation score of lung tissue, MP and MPDNA of BALF was detected after 7, 10 and 14 days of MP infection. MP of BALF was tested by polymerase chain reaction (PCR) test, and MPDNA content was tested by fluorescent quantitative PCR (Realtime FQ PCR).
    Results:Seven days after MP infection models were made, the pathological changes, the positive rate and the content of MPDNA in B group were obviously higher than those in group A, which showed that MP infection models were made successfully. In group C and group F pulmonary inflammation had obvious improvement; group D achieved effective results after 10 and 14 days of infection; group E did not achieve obvious effect. Group C, D and F had significantly more effect than other groups in reducing MPDNA in BALF; the effect of group E was not obvious at 7th and 10th day, but had an advantage at 14th day.
    Conclusions:It shows that Qingzao Jiufei decoction is effective in antiMP, and the efficacy might be correlated with dose and time of administration and infection.

    Objective:To discuss the Yiqi Guben capsule role in regulating the splenic lymphocyte proliferation of lungQi deficiency mice.
    Methods:Totally 96 mice (male and female in half) were equally divided into 6 groups: normal control group, model group, Yiqi Guben capsules highdose group, middledose group and lowdose group, control group of Yupingfeng powder. Apart from the normal group, the mice in the rest of each group were made into lungQi deficiency syndrome models by adopting improved smoked method; tested drugs were given by intragastric administration for five weeks; XTT method was taken to assay the proliferation of splenic lymphocyte stimulated by the LSP or ConA.
    Results:After lungQi deficiency modeling,the function of the splenic B and T lymphocyte proliferation was significantly lower than the normal control group (P<0.001);after the tested drugs were given by intragastric administration, the function of splenic B and T lymphocyte proliferation was significantly increased; compared  with the model group, the splenic lymphocyte proliferation function of control group, of Yupingfeng powder group, of Yiqi Guben capsule highdose group and middledose group was significantly increased(P<0.05),and the increase of Yiqi Guben capsule highdose group was the most significant.
    Conclusions:Yiqi Guben capsule can improve the lymphocyte proliferation activity of lungQi deficiency mice,thereby regulate the immune function.

    Influence of Xiaoban Yushen compound on renal tissue IL13 and CIC in serum of rats with IgA nephropathy

    Objective:To observe the effect of Xiaoban Yushen compound on renal tissue IL13 expression and circulating immune complexes(CIC) serum concentration and to study the mechanism of its treatment for IgA nephropathy.
    Methods:The rats were randomized into normal group,model group,telmisartan group and Xiaoban Yushen compound group, with 10 rats in each group.Bovine serum albumin(BSA) was taken orally to make experimental IgA nephropathy rat model. Xiaoban Yushen compound and telmisartan were given at the eighth week of the experiment for gavage, once daily for five consecutive weeks; at the end of the 12th week, blood was taken to detect immune complexes content, and kidney tissue was taken to detect IL13 expression.
    Results:Compared with the model group, IL13 expression and serum concentration of CIC in Xiaoban Yushen compound group decreased (P<0.01); compared with telmisartan group, IL13 expression and serum CIC concentration in Xiaoban Yushen compound group decreased (P<0.05); both differences had statistic significance.
    Conclusions:Xiaoban Yushen compound plays a role in regulating IL13 expression in renal tissue and serum CIC concentration of IgA nephropathy.

    Objective:To observe the influence of Yiqi Guben capsule on the spleen lymphocyte proliferation function and organ index in the kidneyYang deficiency asthma model mice and to explore its clinical significance.MethodsTotally 96 mice were randomly divided into seven groups: normal control group, asthma group, the kidneyYang deficiency asthma group, kidneyqitonifying pill group, the Yiqi Guben capsule lowdose group, middledose group, highdose group. Apart from the asthma group consisting of 12 mice, the remaining groups each consisted of 14 mice(male and female in half). kidneyYang deficiency asthma groups were treated with Yiqi Guben capsule. Recorded changes in the general condition of the mice in each group during modeling and the period of administration, such as, body weight, body temperature and activity, hair color, diet, breathing frequency, etc. After the experiment, took the spleen and examined the lymphocyte proliferation function; took the kidney, spleen and thymus, and weighed them and calculated the organ coefficient.ReslutsIn the Yiq Guben capsule group and kidneyqitonifying pill group, the mice weight and thymus weight were significantly higher than the kidneyYang deficiency asthma group; in the Yiqi Guben capsule middledose group, the highdose group and kidneyqitonifying pill group, the spleen lymphocyte proliferation by LPS stimulation was significantly higher than the kidneyYang deficiency asthma group.ConclusionYiqi Guben capsule can significantly improve the general state of the kidneyYang deficiency asthma mice, restore immune organ weights, and improve the function of the spleen lymphocyte proliferation.

    Objective:To observe the impact of Yiqi Guben capsule on the splenic lymphocyte in vitro proliferation and organ exponent of spleenQi deficiency asthma mice and study its significance.MethodTotally 96 mice were randomly divided into 7 groups, apart from the asthma group consisting of 12 mice, the remaining groups each consisted of 14 mice(male and female in half). They were normal control group, asthma group, spleenQi deficiency asthma group, Shibeigaoerbao granule group, the Yiqi Guben capsule lowdose group, middledose group and highdose group. Lymphocyte transformation test was used to measure the function of splenic lymphocyte in vitro proliferation; the spleen, thymus, and kidney were taken and weighed, the organ exponent was calculated and the differences were compared between each group.ResultThe mice's thymus weight in the Yiqi Guben capsule lowdose group,middledose group, highdose group and Shibeigaoerbao granule group did not increase significantly; the spleen lymphocyte proliferation stimulated by lipopolysaccharide in Yiqi Guben capsule middledose group, highdose group and Shibergaoerbao granule group was significantly higher than the spleenQi deficiency asthma group(P<0.05).ConclusionYiqi Guben capsule can significantly improve the asthma state of spleenQi deficiency asthmatic mice, and improve the function of spleen lymphocyte proliferation.

    Effect of IGF1 on renal interstitial fibrosis in rats with unilateral ureteral obstruction

    Objective:To investigate the effect of IGF1 on renal interstitial fibrosis in rats with unilatreal ureteral obstruction(UUO).
    Methods:Totally 30 rats were randomly divided into 3 groups, with 10 rats in each group, namely, sham group, model group and IGF1 group. Renal interstitial fibrosis models were made by unilateral ureteral ligation. In the sham group, the abdomen was opened and then stitched; in the model group, physiological saline was intragastrically injected, 0.5 mL a day; in IGF1 group, recombinant human IGF1 was intragastrically injected, 750 mg/(kg·d). At the 14th day after the operation, the rats were anesthetized, and related materials were taken, the kidney function was tested, and   ColⅢ,TGFβ1 and PAI1 expressions in kidneys were observed.
    Results:The expression of BUN,Scr,ColⅢ,TGFβ and PAI1 in the model group were significantly higher than those in sham group, the difference having statistic significance. After the intervention of IGF1, the level of BUN,Scr,ColⅢ,TGFβ and PAI1 in IGF1 group was significantly higher than that in sham and model group, the difference had statistic significance(P<0.05).
    Conclusions:IGF1 plays a role in the pathophysiological process of obstructive nephropathy,especially in the renal interstitial fibrosis.
