Expression and significance analysis of miRNA-126 and runt-associated transcription factor 3 in peripheral blood of children with bronchial asthma

Abstract:

Objective To investigate the expression and significance of miRNA-126 and RUNX3 in children with bronchial asthma.Methods Eighty children with bronchial asthma treated in Henan Children's Hospital were selected as research subjects,and they were divided into acute stage group(n=42) and remission stage group(n=38) according to the clinical progress of the disease. At the same time, 40 healthy children were selected as the control group. The levels of miRNA-126 in peripheral blood of the three groups were detected by qRT-PCR, and the expression level of RUNX3 protein was detected by Western blot; The correlation between miRNA-126 and RUNX3 level in asthmatic groups was analyzed. The diagnostic value of miRNA-126 and RUNX3 in asthma was analyzed by ROC curve.Results Compared with the control group, the expression level of miRNA-126 was significantly increased(P<0.05), while RUNX3 protein was significantly decreased(P<0.05) in both the remission and acute stage groups. The level of miRNA-126 in acute stage group was significantly higher than that in remission group, while the level of RUNX3 protein was significantly lower than that in remission group (P<0.05).The miRNA-126 was significantly negatively correlated with RUNX3 level (P<0.05). The AUCs of miRNA-126 and RUNX3 in diagnosing asthma were respectively 0.933 and 0.847,and the AUCs for the diagnosis of clinical stages of asthma were respectively 0.696 and 0.798.Conclusion The miRNA-126 and RUNX3 may be synergistically involved in the development of bronchial asthma in children, which may serve as a potential auxiliary indicator for the diagnosis and evaluation of asthma
