    Effect and mechanism of oxytocin on maternal aggression in stress model rats during pregnancy period

    Objective To investigate the effect and protective mechanism of different-dose of oxytocin on maternal aggression in stress model rats during pregnancy period.Methods Female Wistar rats(n=40) were randomly divided into four groups named group A(n=10),group B(n=10), group C(n=10) and group D(n=10). During the intervals from 14th day to due day groups(A,B,C) were intraperitoneally injected with 1 mL dilated solution of 0.3, 1.0, 2.0 mg/kg of oxytocin respectively before the forced swimming once a day; meanwhile, group D as control group was given equal volume of normal saline. Some indexes of maternal aggression were tested, such as biting times, keep-down times, attack times, attack duration and latent period, on the 3rd day of parturition .HE staining and immunohistochemical method were used to test changes of oxytocin nuclei injury and oxytocin expression level.Results Regarding biting times group B and C were significantly different from group D(P<0.01);for attack times group C was different from group D(P<0.05);as to attack duration group B and C were different from group D (P<0.05);for attack latent period group B and group C were significantly different from group D(P<0.01). HE staining showed that in supraoptic nuclei(SON) the four groups were significantly different in staining, numbers and density(P<0.01); in paraventricular nuclei(PVN),group C was different from group D(P<0.05),while there was no difference in group A,B and D(P>0.05).Immunohistochemical method showed in SON gray density there was no difference in both group A and D(P>0.05); group C was significantly different from group D(P<0.01); in PVN gray density, group B was significantly different from group D(P<0.01), while there was no difference in group A and D(P>0.05).Conclusion Stress would obviously impair maternal aggression of pregnant rats; different doses of oxytocin could regulate such behavior as influenced by stress and meditate rat brain nuclei oxytocin expression level so that swelling and dissolution of nuclei could be attenuated.

    Objective To observe the effects of different doses of Yiqi Guben capsules on pathology and TGF-β1/Smad signaling pathway in airway remodeling of asthmatic mice.Method The 60 male SPF level BALB/c mice were randomly divided into normal group, model group, dexamethasone intervention(DSMS) group, Tongkang(TKP) group, Yiqi Guben capsules high dose(YQGBH) group and Yiqi Guben capsules low dose(YQGBL) group, with 10 mice in each group. Apart from the normal group, the other groups were sensitized and stimulated by ovalbumin to copy asthma model. On and from the 21st day, each group was given gavage with pure water and medicines 1 h prior to each excitation except the normal group.Observe pathological changes of airway and lung tissues in each group with HE staining, and detect expression of TGF-β1 and P-Smad3 protein in lung tissues with  immunohistochemistry.Result The trachea and lung tissue pathological change degree in model group was statistically higher than that of normal group, model group, DSMS group, TKP group, YQGBH group and YQGBL group according to airway pathological examination(P<0.05);comparing DSMS group with YQGBH group and YQGBL group,the difference was not statistically significant(P>0.05).Compared with normal group,TGF-β1 protein expression levels in model group, DSMS group, TKP group, YQGBH group and YQGBL group statistically increased(P<0.01); compared with model group, TGF-1 protein expression levels statistically decreased in DSMS group, TKP group, YQGBH group and YQGBL group(P<0.01); comparing DSMS group with YQGBH group and YQGBL group,there was no statistical significance(P>0.05);comparing YQGBH group with YQGBL group,the difference was not statistically significant(P>0.05);compared with normal group, the expression levels of P-Smad3 protein increased in DSMS group, TKP group, YQGBH group and YQGBL group(P<0.01);compared with model group, the P-Smad3 protein expression levels decreased in DSMS group, TKP group, YQGBH group and YQGBL group with statiscal significance(P<0.01);the P-Smad3 protein expression level of  DSMS group was even lower than TKP group, YQGBH group and YQGBL group(P<0.01) and the difference was of statistical significance;comaring TKP group with YQGBH group, the difference was not statistically significant(P>0.05);comparing YQGBH group with YQGBL group, there was no statistically significant difference(P>0.05). Conclusion Yiqi Guben capsules can block TGF-β1 intracellular signal transduction by inhibiting excessive expression of TGF-β1 and P-Smad3 in asthma mice, which to a certain extent, reduces asthma airway inflammation in mice and inhibits the occurrence of airway remodeling.

    Effect of scorpion ephedra antispasmodic granules for asthma on p38MAPK and Th1/Th2 cytokines in a mouse model of asthma

    Objective To explore the effect of scorpion ephedra antispasmodic granules for asthma on airway inflammation in asthmatic mice and Th1/Th2 type cytokines changes and its possible mechanism.Methods Totally 24 male mice of only 6 to 8 weeks old were randomly divided into four groups:normal control group, asthma model group, small-dose traditional Chinese medicine group, large-dose Chinese medicine group, 6 mice in each group. Make bronchial asthma model, and the samll-dose traditional Chinese medicine group and large-dose TCM group were given scorpion ephedra antispasmodic granules for asthma to let boiled preparation, 0.72 and 2.89 g daily to make 20 mL/kg aqueous mix solution for stomach irrigation twice a day, for 10 days; normal control group and the asthma model group were given the equal amount of saline. Enzyme linked immune sorbent assay and immunoblotting were used to detect leukocyte mediated-4(IL-4), IL-5, interferon gamma(IFN-γ) content in bronchial alveolar irrigation lavage fluid (BALF) and phosphorylated p38 mitogen activated protein kinase(p38MAPK) changes in the lung tissue, and the pathological changes of inflammatory cells in BALF and lung tissue were observed.Results In BALF of asthmatic model group,inflammatory cell count, IL-4 and IL-5 levels increased and IFN-γlevels decreased; in lung tissues phosphorylated p38MAPK expression level increased, and compared with the normal control group, the difference was statistically significant(P<0.05). The inflammatory cell counts, IL-4 and IL-5 levels decreased and IFN-γ levels increased, in BALF of large-dose Chinese medicine group increased; in lung tissues the phosphorylated p38MAPK expression level reduced, and compared with the asthma model group, the differences were statistically significant(P<0.05). Comparing small with large-dose TCM group, the difference was not statistically significant(P>0.05). Conclusion p38MAPK may be involved in the pathogenesis of bronchial asthma. Scorpion ephedra antispasmodic granules for asthma has a protective effect on asthma in mice, and the mechanism may be related to inhibiting p38MAPK phosphorylation, regulating the imbalance of Th1/Th2 cells and reducing the inflammatory cell infiltration.
