ObjectiveTo observe the regulating effects of drug-containing serum of Li-pi compound preparation on LP,NPY and CCK-8 proteins and genes in rat hypothalamic neuron cells and to explore the mechanism of its treatment for anorexia.MethodsFifty rats were randomly divided into control group, low-, medium- and high-dose Li-pi compound preparation group and zinc sulfate group, with 10 in each group. Control group was given equal volume of drinking water by gavage. Zinc sulfate group was given 20 mg/kg zinc sulfate oral solution. Low-, medium- and high-dose Li-pi compound groups were given 2.4,4.8 and 9.8 g/kg of the compound, respectively. Each group was given gavage once a day for 7 days in a row, and the femoral artery blood was collected on the 7th day after gavage to prepare the drug-containing serum. Rat hypothalamic neuron cells were cultured in vitro and identified by MAP2 immunofluorescence. The hypothalamic neurons were randomly divided into control group, low-, medium- and high-dose Li-pi compound preparation group, and zinc sulfate group by CCK-8, and effect on the activity of hypothalamic neuron cells was assessed at 0 h, 2 h,4 h,6 h,8 h,12 h,24 h,36 h and 48 h. After 24 hours of the corresponding serum, the activity of hypothalamic neurons in each drug-containing serum group began to increase and became different. ELISA was used to detect LP, NPY and CCK-8 content in supernatant of hypothalamic neuron cells; Western Blot and real-time fluorescence quantitative PCR were used to detect the relative expression of LP, NPY and CCK-8 protein and genes in hypothalamic neuronal cells.ResultsThe cells were identified as hypothalamic neuronal cells. Compared with the control group, the LP,NPY and CCK-8 content in supernatant of hypothalamic neuron cells was decreased in the supernatant of hypothalamic neuron cells of low-,medium- and high-dose Li-pi compound preparation, and the differences were statistically significant(P<0.05);the relative expression of LP,NPY and CCK-8 protein in hypothalamic neuron cells in Li-pi compound preparation of medium and high doses was decreased, and the differences were statistically significant(P<0.05); the relative expression of LP gene in hypothalamic neuron cells in high-dose Li-pi compound preparation group was decreased, and the difference was statistically significant(P<0.05); the relative expression of NPY and CCK-8 genes in hypothalamic neuron cells in low-, medium- and high-dose Li-pi compound preparation groups was decreased, and the differences were statistically significant(P<0.05).ConclusionLi-pi compound preparation may improve anorexia-related symptoms by comprehensively regulating the expression of LP, NPY and CCK-8 proteins and genes in hypothalamic neurons, but its effect is negatively correlated with dose.

    Abstract ( 89 )   PDF (835KB) ( 75 )   Save
Blood purification is an important adjunctive therapy for pediatric sepsis.Continuous blood purification(CBP) and plasma exchange(PE) are commonly used models in the treatment of pediatric sepsis. The efficacy of PE in the treatment of sepsis in children is currently not supported by large randomized controlled studies.CBP has the advantages of preventing and treating fluid overload, stabilizing hemodynamics, and continuously replacing or protecting damaged organs,so it is often used as the adjuvant therapy for septic shock and sepsis-associated organ dysfunction(SAOD) in children.High-volume hemofiltration is not recommended for CBP dosage, but there is no consensus on the timing of CBP intervention. In this paper, the research progress in CBP in treating sepsis in children was discussed.

    Abstract ( 96 )   PDF (793KB) ( 217 )   Save
Refractory Mycoplasma pneumoniae pneumonia(RMPP) is a common disease in children. In recent years, its proportion in children′s community-acquired pneumonia is getting higher and higher. With the abuse of antibiotics, the incidence of refractory Mycoplasma pneumoniae pneumonia is increasing.Clinically it is found that RMPP is increasingly resistant to azithromycin, and its treatment is also quite difficult. In traditional Chinese medicine,it is believed that the children are primarily in deficiency of Qi and are more likely to become ill,and the illness progresses rapidly.Based on this,Professor Han Xue puts forward that the main pathogenesis of RMPP is the deficiency of Qi and the complication of evil. Therefore, the treatment should be focused on the defense of positive Qi with both treatment and nourishment being carried out simultaneously.Later, both Yin and Yang should be strengthened and the evil should be treated in order to achieve good clinical effect.

    Abstract ( 51 )   PDF (779KB) ( 24 )   Save
ObjectiveTo investigate the prescription regularity in treating HSP.MethodsThe electronic medical records of 107 HSP child patients in the Affiliated Hospital of Shandong University of Traditional Chinese Medicine from May 2016 to September 2020 were collected, and were screened and analyzed which were used to build a prescription database. Frequency statistics method, association rules and cluster analysis(K-means algorithm+regression models) were used to process data mining. Analyze the prescription regularity of traditional Chinese medicine in treating HSP with the help of TCMISS(V3.0) The property and flavor, frequency, meridian distribution, effect and core drug combination of the core drugs were studied.ResultsThere were 16 Chinese herbs with frequency of usage≥20, and the herbs with high frequency of usage included: honey-fried licorice root, rhizoma atractylodis, moutan bark, semen coicis, golden cypress, lithospermum, etc. There were 23 herb pairs with frequency of usage≥30,and the herb pairs with high frequency of usage included rhizoma atractylodis-golden cypress, rhizoma atractylodis-moutan bark-semen coicis, moutan bark-radix paeoniae rubra, etc. Main taste was bitter and sweet, main drug property was cold, and the main meridian distribution was in lung, spleen, liver and heart. ConclusionThe treatment methods for HSP in our hospital were summarized, such as "bitter and sweet law" and "dispelling latent evil". The main drugs are to clear away heat and toxic materials and promote blood circulation and remove blood stasis, and tonifying drugs are used at the same time.

    Abstract ( 88 )   PDF (776KB) ( 13 )   Save
Wu Jutong advocated syndrome differentiation of triple energizer in his treatise on the Detailed Analysis of Epidemic Warm Diseases, and formed a syndrome differentiation system of febrile diseases with Wei Qi Ying Xue and triple energizer as the core. In Volume Ⅵ, Jieer Nan, through combination with the theory of febrile diseases, he explained his unique views on pediatric diseases and therapeutic drugs. Wu Jutong believed that children's "pure Yang" was "immature Yang is not sufficient, and immature Yin was not developed". They were more likely to be affected by evil and to change, so the treatment and medication should be cautious. Based on Wu Jutong's understanding of the physiological and pathological characteristics of children, this paper explains the medication characteristics of Wu Jutong in pediatrics from seven aspects: careful selection of drugs, careful use of bitter and cold drugs, more sweet and less sour drugs, skillful use of pungent and temperature drugs, good use of pungent and cool drugs, paying attention to the spleen and stomach, and supplementation of diet, in order to provide reference for clinical medication in pediatrics.
